Abstract

This study investigates the synthesis and characterization of isoindoline-1-one γ-lactam derivatives prepared via the reaction of phenyl succinic anhydride with Schiff bases, as well as through the reaction of 2-chlorobenzoyl chloride with imines in the presence of phenyllithium under a nitrogen atmosphere in THF solvent. The reactions were monitored using thin-layer chromatography, and the synthesized compounds were characterized by NMR, IR, and mass spectroscopy, with proposed mechanisms suggested for their formation. The biological effects of two selected compounds (A and B) were evaluated using 30 male white rabbits divided into three groups: a control group and two treated groups receiving compounds A or B orally at a dose of 30 mg/kg/day for 30 days. Blood samples were collected at the end of the experiment to assess kidney-related biochemical and enzymatic parameters, and histological examinations of kidney tissues were performed. The results indicated variation in the effects of the two compounds on renal biochemical markers. Histological findings showed no significant changes in the kidneys of rabbits treated with compound A, whereas treatment with compound B caused noticeable pathological alterations, including cellular enlargement, nuclear atrophy, fibrosis, blood congestion, and inflammatory cell infiltration compared to the control group.
